Prilosec Drug Class and Mechanism

Omeprazole (INN) (pronounced) is a proton pump inhibitor used in the treatment of dyspepsia, peptic ulcer disease (PUD), gastroesophageal reflux disease (GORD/GERD) and Zollinger-Ellison syndrome. It was first marketed by AstraZeneca as the magnesium salt omeprazole magnesium under the trade names Losec and Prilosec, and is now also available from generic manufacturers under various trade names. Omeprazole is one of the most widely prescribed drugs internationally and is available over the counter in some countries. Omeprazole is the racemate (containing a tricoordinated sulfur atom in a pyramidal structure and therefore can exist in equal amounts of both the S and R enantiomers). In the acidic conditions of the stomach, both are converted to achiral products, which reacts with a cysteine group in H+/K+ ATPase, hereby destroying the ability of the parietal cells to produce gastric acid.

Prilosec Uses

Prilosec (Omeprazole) is used to treat various acid-related stomach and/or throat (esophagus) problems (e.g., GERD, ulcers, erosive esophagitis, or Zollinger-Ellison Syndrome). It works by blocking the production of acid in the stomach. When using Prilosec to treat certain ulcers, it may be prescribed along with certain antibiotics (e.g., amoxicillin, clarithromycin). Omeprazole is available as tablets and capsules (containing omeprazole or omeprazole magnesium) in strengths of 10 mg, 20 mg, and in some markets 40 mg; and as a powder (omeprazole sodium) for intravenous injection.

Prilosec Side Effects

Headache, diarrhea, gas, or constipation, may occur. If any of these effects persist or worsen, notify your doctor. Tell your doctor immediately if any of these serious side effects occur: rash, stomach/abdominal pain, back pain, unusual tiredness, dizziness, vomiting. Tell your doctor immediately if any of these unlikely but serious side effects occur: chest pain, dark urine, yellowing eyes or skin. An allergic reaction to Prilosec is unlikely, but seek immediate medical attention if it occurs. Symptoms of an allergic reaction include: rash, itching, swelling, trouble breathing. Prilosec Pregnancy

Pregnancy Category C There are no adequate and well-controlled studies on the use of omeprazole in pregnant women. The vast majority of reported experience with omeprazole during human pregnancy is first trimester exposure and the duration of use is rarely specified, e.g., intermittent vs. chronic. An expert review of published data on experiences with omeprazole use during pregnancy by TERIS – the Teratogen Information System – concluded that therapeutic doses during pregnancy are unlikely to pose a substantial teratogenic risk

Prilosec Overdose

Reports have been received of overdosage with omeprazole in humans. Doses ranged up to 2400 mg (120 times the usual recommended clinical dose). Manifestations were variable, but included confusion, drowsiness, blurred vision, tachycardia, nausea, vomiting, diaphoresis, flushing, headache, dry mouth, and other adverse reactions similar to those seen in normal clinical experience.
